The City of Prague has 4 wheelchair accessible taxi cabs which can accommodate both manual and powered wheelchairs. The accessible taxi vans have a lowered floor and are equipped for rear (lift gate) entry. The driver will secure your wheelchair using standard-style straps, and can attach a seatbelt for your use.
In my experience, at least one hour’s notice should be provided to secure an accessible taxi. As is true of all cities, response times vary, so making reservations as far in advance as possible is recommended. Accessible taxis operate 24 hours a day in Prague, and are provided by Centrum Mobility, a government-supported service:

Taxi meters operate at the following rates:
Flag Drop — 30 CZK (~$1.20 USD)
Per kilometer — 24.90 CZK
Waiting time (per minute) — 4 CZK
Night surcharge (9 p.m. to 6 a.m.) — +30%
Trip minimum — 250 CZK (~$10.00 USD)
Flat Rate (Airport to/from Prague) — 680 CZK
The taxis are among the most inexpensive I have found in Europe, and the drivers are very friendly. While I cannot confirm that all drivers of the accessible taxis speak English, my drivers did.
